Development of protein-protein interactions have been recalcitrant to small molecule modulation and yet lend themselves easily to interruption with antibodies. The search for an agent that shares the characteristics of cell permeability of small molecules and specificity of antibodies has lead to the development of novel peptides (cyclic/stapled peptides). A successful story of interrogating the p53 pathway will be discussed.
